基于ATR-FTIR结合UPLC-DAD技术鉴别百合及其类似品(1)

     摘要:目的 基于傅里叶变换衰减全反射红外光谱(ATR-FTIR)结合超高效液相色谱-二极管阵列检测器(UPLC-DAD)技术快速区分不同品种百合药材粉末。方法 采用ATR-FTIR采集百合样品红外光谱数据并导入Simca软件,建立正交偏最小二乘判别分析(OPLS-DA)模型,在此基础上,联用UPLC-DAD技术测定正品百合、伪品金百合及微波干燥卷丹中王百合苷A含量,分析其差异性。结果 OPLS-DA二维得分图及层次聚类分析图对卷丹与百合、卷丹与伪品金百合、热风干燥卷丹与微波干燥卷丹進行了良好的分类。卷丹的王百合苷A含量为2.09~3.28 mg/g,百合的王百合苷A含量为1.36~1.77 mg/g,伪品金百合的王百合苷A含量为1.03~1.05 mg/g,微波干燥卷丹的王百合苷A含量为3.92~4.01 mg/g。卷丹和百合中的王百合苷A含量高于伪品金百合,微波干燥卷丹的王百合苷A含量高于普通热风干燥卷丹。结论 本研究建立的方法操作简单、方便、快速,可为百合饮片、百合药膳产品开发及相关药材的质量控制提供借鉴。

    Abstract: Objective To quickly distinguish different kinds of Lilii Bulbus powder based on ATR-FTIR and UPLC-DAD. Methods The infrared spectral data of samples were collected by ATR-FTIR, and the data was imported into Simca software to establish the orthogonal partial least squares discriminant analysis (OPLS-DA) model. On this basis, UPLC-DAD was used to determine the content of regaloside A in genuine Lilii Bulbus and Lilium trompeten (counterfeit) and microwave dried Lilium lancifolium Thunb., and the differences among different samples were analyzed. Results Two-dimensional score map of OPLS-DA and hierarchical cluster analysis were used to classify the three groups of Lilium lancifolium Thunb. and Lilium broumii F.E. Brown var.viridulum Baker, Lilium lancifolium Thunb. and Lilium trompeten (counterfeit), hot-air dried Lilium lancifolium Thunb. and microwave dried Lilium lancifolium Thunb. The content of regaloside A in Lilium lancifolium Thunb. was 2.09–3.28 mg/g, 1.36–1.77 mg/g in Lilium broumii F.E.
